Thượng Xuân Đường là một tiệm thuốc danh tiếng. Ông chủ họ Đỗ không có con trai, chỉ có duy nhất một đứa con gái là Đỗ Giai Kỳ. Tuy Giai Kỳ hứng thú với y thuật và rất hiếu học, nhưng cha cô không trông mong cô có thể kế thừa gia nghiệp.

Giai Kỳ may mắn gặp được một thần y đã thoái ẩn, được ông chỉ dạy y thuật, đồng thời hóa thù thành bạn với Nhất Nguyên. Mặt khác, em trai Trang Kế Tổ là Trang Kế Tông thầm yêu Giai Kỳ nên từ chối tình yêu của Hứa Quân Ước con gái một tướng quan quân phiệt, Quân Ước cũng vì vậy xem Giai Kỳ là cái gai trong mắt.
